Application of Ultrafiltration and Ion Exchange Separation Technology for Lysozyme Separation and Extraction

In this study, the fermentation broth of recombinant Pichia pastoris strain ncy-2 was studied. After pretreatment, separation, and purification, lysozyme optimized using biofilm ion exchange separation. Finally, dry enzyme powder prepared by concentrating vacuum drying. The removal rate bacterial cells 99.99% when centrifuged at low temperature. optimum conditions were: transmembrane pressure 0.20 MPa, pH 6.5, 96.6% yield lysozyme, activity 2612.1 u/mg, which 1.78 times higher than that original enzyme; D152 resin used for adsorption elution. Process were optimized: volume ratio to liquid 15%; time 4 h; concentration NaCl 1.0 mol/L; recovery 95.67%; 3879.6 u/mL; purification multiple 0.5, 3.1 activity. 12,573.6 had an inhibitory effect on microsphere lysozyme. Its enzymatic properties almost same as those natural demonstrated good application prospects production potential.
